pP{BeloBAC}



pP{BeloBAC} is a derivative of pBeloBAC11, a vector that is maintained at about one copy per cell. The low copy number greatly increases the stability of cloned repetitive DNA by eliminating the opportunity for inter-molecular recombination events. The tradeoff is that DNA yields are very low. This vector should be grown in a strain that does not already harbor a BAC; we use STBL cells (Gibco/Life Technologies). This derivative has P element ends and some unique cloning sites, but no marker for transformation (I thought about putting one in, but BAC vectors are such a pain to deal with, I just didn't get to it).
